(1)藥效學(xué):本品具有鎮(zhèn)痛、抗炎及解熱作用。本品通過抑制環(huán)氧酶使組織局部前列腺素的合成減少及抑制白細(xì)胞的趨化性和溶酶體酶的釋放而起到藥理作用。本品治療關(guān)節(jié)炎時的鎮(zhèn)痛、消腫等療效與吲哚美辛、阿司匹林、萘普生相似。但由于本品抑制環(huán)氧酶-2所需的濃度高于抑制環(huán)氧酶-1的濃度，因此胃腸道的不良反應(yīng)較多。

(2)藥動學(xué):口服吸收好。食物可降低吸收速度，但不影響吸收總量。血漿蛋白結(jié)合率高達(dá)90%以上。經(jīng)肝臟代謝。半衰期平均為50小時(30~86小時)，腎功能不全患者半衰期延長。由于半衰期較長，一次給藥即可維持24小時的血藥濃度相對穩(wěn)定，多次給藥易致蓄積。一次服藥20mg，3~5小時血藥濃度達(dá)峰值，血藥有效濃度為1.5~2μg/ml。血藥穩(wěn)定濃度在開始治療后7~12天方能達(dá)到。66%自腎臟排泄，33%自糞便排泄，內(nèi)有<5%為原形物。
